摘 要:目的 探讨顺铂 (DDP)加热后对肝癌细胞的毒性 ,温度与DDP杀伤力的关系及其作用机制。方法 采用MTT法、台盼蓝排染法以及流式细胞术分析法。结果 DDP对肿瘤细胞有明显的杀伤力 ,其IC50 值为 0 .5 7μg/ml。当DDP( 2 .0 μg/ml)温度为 45℃时 ,药物作用 2 4h后 ,细胞总数分别是对照组、45℃单热组、单药组的 12 .3 %、2 4.5 %和 44.0 % ,提示一次性加热DDP至 41℃以上可明显提高其抗癌能力。结论 加热增强了DDP的细胞毒性 ,两者之间有着协同增敏作用 。Objective To determine the inhibition effect of heating DDP on Hca F cell lines,and the relationship between temperature and medication effect as well as its antitumor mechanism.Methods The relationship between temperature and cytotoxicity of DDP was observed by typanblue assay?MTT and flow cytometry.Results DDP had obvious cytotoxicity to Hca cell line growth,and the IC 50 value of Hca Fcell lines to DDP was 0.57 μg/ml.At DDP(2.0 μg/ml)45 ℃,for 24 hours the overall cell numbers was 12.3% of the control?24.5% of heating alone and 44.0% of drug alone respectively.Heating DDP improved the effect of cell growth inhibition obviously.Conclusion Healing may enhance the cytotoxicity of DDP.G 0/G 1 phase cells arrested is one of their main antitumor mechanisms.
