环孢霉素A、他莫昔酚对耐药细胞系K562/DOX多药耐药逆转作用的研究

摘　　要：目的　寻找有效的多药耐药联合逆转剂。方法　以他莫昔酚与环孢霉素A作为耐药逆转剂 ,采用MTT法进行体外药物敏感试验 ,观察他莫昔酚与环孢霉素A对多药耐药白血病细胞株K 5 6 2 /DOX的药物敏感性的影响。结果　他莫昔酚与环孢霉素A均可增强阿霉素 (DOX)对K 5 6 2 /DOX的细胞毒作用 ,两者有联合协同作用 ,逆转倍数为单用药的 9倍 ,超过单用 2倍剂量的逆转效果。结论　他莫昔酚与环孢霉素A都是有效的多药耐药逆转剂 。Objective To develop effective combination of drug resistance modulators.Methods The effects of tamoxifen and Cydosporine A on drug sensitivity in multidrug resistant human leukemic cell line K562/DOX were studied by MTT colorimetric assay.Results Both of Tamoxifen and Cydosporine A could enhance the cytotoxicity of doxorubicin(DOX) to K562/DOX.The reversal effectiveness of the drug combination was 9 times as much as that of each drug alone.Conclusion Tamoxifen and Cydosporine A are all effective reversal agent of multidrug resistance.The combination of both has a synergistic interaction.
