摘 要:目的制造一种新的大鼠痴呆模型 ,考察模型大鼠学习、记忆和M受体密度的变化 ,以及知母有效成分知母皂甙元 (ZMS)的影响。 方法单侧基底核内联合注射 β -淀粉样肽 1- 40片段 (Aβ1-4 0 )和兴奋性氨基酸建立大鼠痴呆模型 ,然后将模型动物分为假手术组、模型组、阳性对照组和ZMS三个剂量组。用避暗法及跳台法测定动物的学习、记忆功能 ,用放射配基结合分析法测定脑M受体密度。 结果 模型动物造型 45d后 ,避暗法及跳台法实验错误次数增加 ,潜伏期缩短 ,脑内M受体密度降低。平行的模型动物喂服知母皂甙元 45d后 ,上述指标明显改善。结论用单侧脑内定位注射Aβ和兴奋性氨基酸成功地制造了一种新的痴呆模型 ,ZMS能明显改善其学习、记忆功能并提高其M受体密度。Objective To produce a new type of dementia model so as to examine the functional change of learning and memory, muscarinic acetylcholine receptor density, and effect of ZMS-active component of Zhimu. Methods A combined injection of Aβ 1-40 and excitatory amino acid to the right nucleus basalis was used to produce the model. The model animals were divided into sham operation, model, positive control group and three dosage groups of ZMS. The learning and memory changes were detected by the step-down and step-through tests, and the muscarinic receptor density was determined by 3H-QNB binding tests. Results 45d after injection, the learning and memory ability as well as the muscarinic receptor density were decreased significantly. The parallel models treated with daily oral administration of ZMS for 45d improved in learning and memory ability and increased in muscarinic receptor density. Conclusion Combined injection of Aβ 1-40 and excitatory amino acid to the right nucleus basalis successfully produced a new dementia model. ZMS can significantly improve the learning and memory ability and increase the brain muscarinic receptor density of the model.
