摘  要:目的:研究全反式维甲酸alltransretinoicacid,ATRA对肺腺癌细胞株SPCA1细胞的增殖及其表面粘附分子表达的影响。方法:使用1×10-6mol/L、1×10-5mol/L和2×10-5mol/L的ATRA及IL8、TNFα处理SPCA1细胞并培养72h后,用MTT方法观察细胞的增殖,间接免疫荧光法和流式细胞仪分析SPCA1细胞表面粘附分子的表达。结果:3种不同浓度ATRA都能抑制SPCA1细胞的增殖,抑制率分别为12.1%、30.5%、62.5%,且在浓度为1×10-6mol/L和1×10-5mol/L时与IL8和TNFα具有协同作用。3种不同浓度ATRA还可抑制SPCA1细胞表面CD11α、CD62E和CD62L粘附分子的表达;在浓度为1×10-6mol/L时抑制作用最明显,抑制率分别为84.6%、60.5%、62.0%;但对CD18和CD54分子的表达无明显作用。结论:ATRA能抑制SPCA1细胞增殖,降低其粘附分子的表达,提示ATRA对肺腺癌细胞的生长及转移可能有抑制作用。Objectives:To investigate the in hibitory effects of all-trans retinoic acid(ATRA)on the proliferation of lung adenoma cell line SPCA-1cells a nd the expression of adhesive molecu les on t he surface of the cells.Methods :After incubation with IL-8and TNF-αfor 7 2h,the effect of ATRA with concentrations of 1×10 -6 ,1×10 -5 ,and 2×10 -5 mol /L respectively on the prolifera tion of SPCA-1cells was analyzed by MTT method.The expression of adhesiv e molecules on the surface of SPCA-1cell s was determined by flow cytometry.Results:The proliferation of SPCA-1cells was inhibited significantly a fter incubation with three differen t concentratio ns of ATRA.And the inh ibition rate was12.1%,30.5%,and 62.5%re spectively.1×10 -6 and 1×10 -5 mol /L ATRA coordinating with IL-8an d TNF-αcan inhibit the proliferati on of SPCA-1cells.The expressions of CD11α,CD62E,and CD62L can be si gnificantl y inhibited by three different concentrations o f ATRA.1×10 -6 mol /L ATRA was the most,and the inhib itory rate was 84.6%,60.5 %,62.0%respectively.but there were no effects on the expression of CD1 8and CD54.Conclusion:ATRA can inhibit the proliferation of SPC A-1cells a nd reduce the expressions of adhesive molecules on the surface of SPCA-1cells .ATRA may be used in the control of the proliferation and metastasis of lung ad enoma cancer.
