摘 要:目的 评价阿仑膦酸钠、埃本膦酸钠、酪蛋白磷酸肽对去势雌性大鼠骨质疏松症的预防作用。方法 5 9周龄去势SD雌性大鼠随机分为 4组 ,即阿仑膦酸钠组、埃本磷酸钠组、酪蛋白磷酸肽组和对照组。分别给予相应干预药物持续处理 6个月后测定骨密度及灰重 ,并进行骨组织形态计量学及骨生物力学研究。结果 连续给药 6个月后 ,全身、腰椎和股骨头骨密度分析显示埃本膦酸钠组显著高于对照组 (P <0 0 1,P <0 0 1,P <0 0 5 ) ;胸椎、胫骨骨小梁面积测量也以埃本膦酸钠组为优 (P <0 0 1,P <0 0 1) ;股骨灰重检测表明埃本膦酸钠组也明显高于对照组 (P <0 0 5 )。阿仑磷酸钠组的骨小梁面积显著高于对照组 (P <0 0 1,P <0 0 1) ,但消化道毒性反应较重。酪蛋白磷酸肽组除全身骨密度外 (P <0 0 5 ) ,其它所测参数与对照组比较无明显差异。结论 埃本磷酸钠是一种较好的预防绝经后骨质疏松症的药物。对于阿仑膦酸钠的使用 ,建议可通过改变药物剂型和给药途径来完善。酪蛋白磷酸肽作为骨质疏松症的单一预防用药尚有局限性 。Objective To evaluate the preventive effect of alendronate,ibandronate and casein phosphopeptides Ⅲ(CPP\|Ⅲ) on osteoporosis in ovariectomized rats. Methods\ 59 week\|old SD female rats were ovariectomized and divided into four groups randomly:alendronate,ibandronate,CPP\|Ⅲ and control groups.The treatment groups were fed with the above druge for consecutive six months.Later,bone mineral density(BMD) and ash weights were measured,and bone histomorphometry was performed. Results\ The best BMDs of total body,lumbar spine and femoral head were found in ibandronate group,and they were significantly higher than those in the control group( P <0\^01, P <0\^01, P <0\^05,respectively);larger trabecular area of thoracic vertebra and tibia( P <0\^01, P <0\^01,respectively)and higher ash weight of femur were also obtainedin the ibandronate group( P <0\^05).The trabecular area in alendronate group was much larger than that in control group( P <0\^01, P <0\^01),but the toxicity to alimentary tract was severe.No significantly higher parameters were found in CPP\|Ⅲ group except the BMD of total bady( P <0\^05). Conclusion\ Bisphosphonates are now well established as efficacious antiresorptive agents for the prevention of osteoporosis.In particular,ibandronate has been proved to be able to increase bone mass and improve bone physical condition with slight toxicity.Alendronate may be improved by altering dosage form and mode of administration.CPP\|Ⅲ still has limited use,but may be a good adjuvant for prevention of osteoporosis.
