摘  要:从团头鲂(Megalobrama amblycephala)和广东鲂(Megalobrama hoffmanni)的脑垂体组织提取总RNA,采用3’RACE-PCR的方法,从中扩增出编码两种鲂生长激素(growth hormone,GH)成熟肽的cDNA序列,克隆到pGEM-T EasyVector载体上。克隆的团头鲂和广东鲂GHcDNA均包括编码188个氨基酸残基的GH成熟肽序列和3’端的非翻译区,但不含信号肽序列和5’端非编码区。序列分析结果表明,团头鲂和广东鲂GH的碱基序列和推测的氨基酸序列的同源性分别为99%和100%。Total RNAs was isolated from the pituitary of Megalobrama hoffmanni and Megalobrama amblycephala. The cDNAs encoding two species of gurnards GH mature peptides together with 3' end untranslated regions were amplified by 3'RACE-PCR strategy using isolated total RNAs as template. The amplified cDNA fragments were inserted into pGEM-T Easy Vector. The cloned cDNAs in- duded no signal peptides sequence. The GH mature peptides of both gurnards were composed of 188 amino acid residues. The consistecy of the two gurnards nucleotide sequences of mature GH peptides and deduced amino acid sequences were 99% and 100% respecfively.
