癌基因MDM2mRNA在儿童急性淋巴细胞白血病的过度表达(附32例报告)

摘　　要：应用逆转录聚合酶链反应方法检测 32例急性淋巴细胞白血病 (AL L )患儿的外周血单个核细胞MDM2 m RNA表达 ,以 β- actin为内参照 ,对结果进行半定量。结果 MDM2 m RNA在儿童 AL L 中过度表达率明显高于正常对照组 ;在不良预后组、高白细胞性白血病组中的表达分别高于一般预后组、非高白细胞白血病组。认为 MDM2过度表达在儿童 AL L 发病中起有重要作用 ,并与儿童 AL L 的不良预后。The level of MDM2mRNA expression in peripheral blood from 32 patients with acute lymphoblastic leukemia were evaluated by RT PCR. Peripheral blood specimens from 22 healthy children were normal controls. MDM2mRNA over expressed in patients then in healthy. We divided these patients into several subgroups including T ALL and Non T ALL, L 1 and L 2 , hyperleukocytic acute leukemia(HAL) and nonhyperleukocytic acute leukemia(NHAL), Good prognosis and poor outcome,and compared the levels of MDM2mRNA of these subgroups respectively. MDM2mRNA over expressed dramatically in children with ALL than that in healthy children. Unfavorable prognositic patients showed a striking over expression of MDM2 compared to good outcome subgroup. Although we did not find significant difference of MDM2 expression between T ALL and Non T ALL (P=0.7345) as well as L 1 and L 2 subgroup (P=0.8923), We detected MDM2mRNA overexpression dramatically in HAL compared with NHAL subgroup (P=0.0099). These findings suggested that MDM2mRNA over expression plays animportant role in children ALL. Furthermore, MDM2mRNA over expression is associated with HALL
