摘 要:本文以感病自交系 Mo17、掖 10 7和抗病自交系黄早四为材料 ,研究了玉米矮花叶病毒(MDMV)和 MDMV辅助成份—蛋白酶 (HC- Pro)在叶片中的积累动态和细胞内定位。结果表明 ,在接种后 3d,MDMV就在掖 10 7和 Mo17接种叶的上位叶积累到了相当高的程度 ,6d后达到高峰 ;而MDMV在黄早四中的浓度一直低于在掖 10 7和 Mo17中的浓度。在接种掖 10 7和 Mo176d后 ,MDMV HC- Pro在上位叶中的积累到达高峰 ,而在黄早四植株中 MDMV HC- Pro直到第 9d才到最高 ,而且浓度一直低于感病自交系掖 10 7和 Mo17。麦二叉蚜从发病植株上获毒后的传毒效率变化与MDMV、MDMV HC- Pro在叶片中的积累动态一致。通过胶体金标记对 MDMV HC- Pro进行免疫定位 ,发现在细胞质中、与质膜相联系的风轮状内含体、片层凝集状内含体、细胞质高密度物质和病毒粒子周围有金标记 ,说明在这些部位有 MDMV HC- Pro的存在。The time course and sub cellular location of Maize dwarf mosaic virus (MDMV) and its helper component proteinase (HC Pro) in infected maize leaves were determined. Among the maize lines tested in the experiment, Ye107 and Mo17, were highly susceptible to MDMV, while Huangzao 4 was resistant. The results showed that the concentration of MDMV was higher in the upper leaves of inoculated Ye107 and Mo17 3 days after inoculation, and reached the maximum level 6 days after inoculation, while the concentration of MDMV in Huangzao 4 was always lower than that in Ye107 and Mo17. The level of MDMV HC Pro in the upper leaves just above the inoculated leaves of Ye107 and Mo17 reached maximum 6 days after inoculation, but in the corresponding leaves of Huangzao 4, it reached maximum 9 days after inoculation and kept lower thereafter. The changes of efficiency of MDMV transmission aphids were consistent with the level of MDMV and MDMV HC Pro in the leaves. The gold label was observed on pinwheel inclusion, laminated inclusion and virus particles in the cytoplasm, which suggested that MDMV HC Pro was present in these locations.
