摘  要:目的分析帕金森病 (PD)小鼠模型中脑黑质多巴胺转运蛋白 (DAT)基因转录状况。 方法用 1-甲基 - 4-苯基 - 1,2 ,3,6 -四氢吡啶 (MPTP)腹腔注射C5 7BL小鼠 ,制作PD鼠模型 ,分 10、18d两组 ;用相同体积生理盐水腹腔注射作为正常对照。以Dot-Blot方法检测小鼠中脑黑质部位DAT基因转录。 结果与正常对照相比 ,MPTP组小鼠行为异常 ,中脑黑质部位TH(+ )细胞减少 ,DATmRNA转录降低 (P <0 .0 5 )。正常对照组中小脑部位DATmRNA转录低于黑质部位 (P <0 .0 5 )。 结论多巴胺能神经元及其DATmRNA转录量随PD的发生而减少或降低。DATmRNA转录量可反映残存多巴胺神经元量或功能状况 ,反映PD的状况 。Objective To analyse the transcription of DATmRNA in the substantia nigra of Parkinsonism animal models. Methods To make PD mouse models, C57BL mice were treated with 1-methy1-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) hydrochloride (35mg/kg) and they were divided into two 10d and 18d groups. Another group of mice were treated with the same volume of sodium chloride as normal controls. Then the levels of DATmRNA were detected by the Dot-blot method. Results As compared with the normal controls, the MPTP-treated mice had abnormal behaviour. The positive cells of TH were diminished and the expression of DAT gene was significantly decreased (P<0.05) in the substantia nigra. In normal controls, the expression of DAT gene in the substantia nigra was higher than that in the cerebellum. Conclusion The dopaminergic neurons and the transcription of DATmRNA were reduced along with the occurence of Parkinson's disease. The result suggested that the transcription of DATmRNA might express the amount and the function of surviving dopaminergic neurons and the state of Parkinson's disease. The cells with a higher transcription of DATmRNA were more sensitive to MPTP.
