奥曲肽对MCF-7人乳腺癌细胞株生长及凋亡的影响

摘　　要：目的：研究生长抑素类似物奥曲肽（ＯＣＴ）对乳腺癌细胞生长及凋亡的影响。方法：将ＯＣＴ作用于体外培养的雌激素受体（ＥＲ）阳性人乳腺癌细胞株ＭＣＦ－７，应用ＭＴＴ比色法分析细胞生长抑制作用，流式细胞术测定细胞周期分布和凋亡率，光镜、电镜观察细胞形态和超微结构。结果：ＯＣＴ可抑制ＭＣＦ－７细胞生长、抑制细胞周期，并可诱导细胞凋亡。结论：ＯＣＴ　对人乳腺癌细胞增殖具有抑制作用，其机制与诱导肿瘤细胞凋亡有关。Objective: To study the effect of somatostatin analogue octreotide (OCT)on growth and apoptosis in breast cancer cell. Methods:Estrogen receptor(ER)-positive human bresast carcinoma cell line MCF-7 was treated with OCT in vitro. Cell proliferation was evaluated by MTT assay,distribution of cell cycle and rate of apoptosis were determined by flow cytometry,and apoptosis was confirmed by morphology and ultrastructural alternations.Results:OCTcould inhibit the growth,arrest the cell cycle, and induce the apoptosis in MCF-7 cells. Conclusion:OCT possessed an inhibitory effect on proliferation of breast cancer cells,and the me chanism for antitumor was associated with induction of apoptosis.
