摘 要:目的 :研究大鼠切口疼痛模型中鞘内 (it)新斯的明 (NEO)、吗啡和NEO加吗啡镇痛对脊髓一氧化氮合酶(NOS)活性、一氧化氮 (NO) /环鸟苷酸 (cGMP)含量的影响。方法 :雄性SD大鼠 12 8只 ,随机均分为 8组 :(1)假手术组 ,(2 )术前 30minit0 .9%氯化钠 2 0 μl(l组 ,对照组 ) ,(3)术后 (术后NEO治疗组 )和 (4)术前 (术前NEO治疗组 )30minitNEO 10 μg组 ,(5 )术后 (术后吗啡治疗组 )和 (6 )术前 (术前吗啡治疗组 ) 30minit吗啡 5 μg组 ,以及 (7)术后(术后NEO加吗啡治疗组 )和 (8)术前 (术前NEO加吗啡治疗组 ) 30minitNEO 5 μg加吗啡 2 .5 μg组。累积疼痛评分确定疼痛行为。分光光度法测定脊髓NOS活性、NO产量 ;放射免疫法测定cGMP含量。结果 :术后NEO治疗、术前NEO治疗、术后吗啡治疗、术前吗啡治疗、术后NEO加吗啡治疗和术前NEO加吗啡治疗各组大鼠的累积评分均明显低于对照组 (P均 <0 .0 1)。在对照组 ,脊髓的NOS活性、NO产量和cGMP含量均显著高于假手术组 (P均 <0 .0 1)。术前NEO治疗、术前吗啡治疗和术前NEO加吗啡治疗组的脊髓NOS活性、NO产量和cGMP含量均较对照组明显降低 (P <0 .0 5或P <0 .0 1)。结论 :在大鼠切口疼痛模型中 ,术前itNEO。Aim: To study the effects of intrathecal (it) neostigmine, morphine and neostigmine plus morphine on nitric oxide (NO) synthase(NOS )activity, NO/cyclic 3',5'-monophosphate (cGMP) content of the spinal cord in a rat model of incisional pain. Methods:128 male SD rats were divided randomly into eight groups. (1) Sham group received it 0.9%NaCl 20μl and 30min later inhaled 1.4% isoflurane for 5min but no incision was made. (2) Control group received it 0.9% NaCl 20μl 30min before incision. The rats in (3) post-incisional neostigmine group and (4) pre incisional neostigmine group received it administration of neostigmine 10μg either 30min after or before incision respectively. The rats in (5) post incisional morphine group and (6) pre incisional morphine group received it administration of morphine 5μg either 30min after or before incision respectively. The rats in (7) post incisional neostigmine plus morphine group and (8) pre incisional neostigmine plus morphine group received it administration of neostigmine 5μg plus morphine 2.5μg either 30min after or before incision respectively. A cumulative pain score was utilized to assess pain behavior. NOS activity, NO production and cGMP content were measured by spectrophotometry and radioimmunoassy. Results: Cumulative pain score in treatment groups were significantly lower than that in control group ( P <0.01). NOS activity, NO production as well as cGMP content of the spinal cord increased significantly in control group as compared with those in sham group ( P <0.01). NOS activity, NO production and cGMP content of the spinal cord in pre incisional neostigmine group, pre incisional morphine group and pre incisional neostigmine plus morphine group were significantly lower than those in the control group ( P <0.05 or P <0.01). Conclusion: In a rat model of incisional pain, antinociceptive effect of the intrathecal administration of neostigmine, morphine, neostigmine plus morphine may be associated with blockade of the NO/cGMP signaling pa
