摘  要:目的 探讨肝癌癌变过程中雄激素受体 (AR)表达的动态变化和病理意义。方法 以二甲氨基偶氮苯 ( 3′ Me DAB)建立大鼠诱发性肝癌模型 ,将 6 0只雄鼠分为 2组 :诱癌组 (n =35 ) ,以 3′ Me DAB诱癌 ,对照组 (n =2 5 ) ,以精制大米喂服。放射配基结合技术检测各实验期大鼠肝脏AR的定量表达。结果 诱癌组大鼠肝重 /体重比值在第 4、5个月时大幅度上升 (P <0 .0 5 )。诱癌组雄激素受体定量以癌变启动期 (第 1个月 )和成癌期 (第 5个月 )最高 ,第 2个月最低 ,细胞核AR分别为 ( 5 6 .2 8± 9.48) ,( 5 9.49± 4.42 )和 ( 33.5 2± 3.2 4)fmol/mg蛋白质。对照组大鼠的生理性AR表达与年龄依赖的性活动能力一致。诱发癌瘤受体定量以肝细胞型肝癌最高 (P <0 .0 5 ) ,且癌瘤组织高于癌周组织 (P <0 .0 5 )。结论 实验性肝癌癌变特定阶段雄激素受体表达活跃 。Objective To investigate dynamic change and the pathological meanings of androgen receptor(AR) expression in hepatocarcinogenesis.Methods 60 male SD rats were divided into two groups:cancer-induced group(3′-Me-DAB),control group(rice only),to establish rat model of chemically-induced hepatocarcinogenesis.The quantity of AR in liver was detected with Radio-ligand Binding Assay at various experimental stages.Results The ratio of liver to body weight rised rapidly at the 4th and 5th month in cancer-induced group.The level of AR in cancer-induced group was the highest at early stage of cancer induction(the first month)and occuring-tumor stage(the 5th month),(56.28±9.48) fmol/mg·protein and (59.49±4.42) fmol/mg·protein,respectively,the lowest at the 2th month(33.52±3.24) fmol/mg.protein.The physiological expression of AR in control group was in accordance with age-dependent sex hormone activity.Of different histological type of induced tumor,AR concentration of hepatocellular carcinoma was the highest(P<0.05),the level of AR in tumor tissue was sigificantly higher than that in surrounding tumor tissue(P<0.05).Conclusion Expression of AR in special stage of hepatocarcinogenesis is active,AR may have a liver cell proliferation-promotion effect.
