摘  要:目的 :为了研究酪氨酸激酶抑制剂 Herbim ycin A(HMA)的作用。方法 :采用不同病期的慢性粒细胞白血病 (CML)细胞和 K5 6 2细胞株进行体外培养 ,加入不同浓度的 HMA及 α-干扰素。结果 :HMA或 α-干扰素单独应用能明显抑制 CML 细胞增殖 ,但不能诱导 CML 细胞凋亡 ,两者联合应用能明显促进 CML 细胞凋亡 ,且能获得明显的遗传学缓解。当加入外源性巯基化合物阻断 HMA与酪氨酸激酶结合时可完全消除这种作用。结论 :HMA联合α-干扰素在 CML的治疗如化疗或自身骨髓移植的体外净化中具有应用价值。Objective:In order to research the effect combinating herbimycinA (HMA).Method:An inhibitor of tyrosine kinase with α IFN on CML cells,cells of CML and normal marrow were cultured in vitro to observe the effect of the induction kill or apoptosis by using conbination of HMA and α IFN. Result:HMA or α IFN could inhibit the proliferation of CML cell but not significantly induce apoptosis of CML cell when using single.However,HMA significantly improved apoptosis when combined with α IFN,and the relief of cytogenetics was obtained obviously. The effect will be completely cut off when sulifhydryl compound was added into the culture to conjugate HMA.Conclusion:That the combination of HMA and α IFN could be used in the treatment of CML as a chemotherapeutic agent or puring approach of auto bone marrow transplantation in vitro.
