肝硬化大鼠全胃肠外营养时一氧化氮和一氧化氮合酶活性的变化及意义

摘  要:目的 观察一氧化氮 (NO)、一氧化氮合酶 (NOS)活性在肝硬化大鼠全胃肠外营养 (TPN)时的变化 ,并探讨其意义。方法 Wistar大鼠分为 3组 :正常组 8只 ,肝硬化对照组 6只 ,肝硬化TPN组 7只。测定血清NO浓度、肝脏NO含量和肝脏NOS比活性 ,观察肝脏 β NADPH黄递酶组化染色。 结果 除TPN组一只大鼠因输液过快 ,肺水肿死亡外 ,其余大鼠均成活。血清NO浓度、肝脏NOS比活性 ,肝硬化对照组比正常组升高明显 (P <0 .0 5 ) ,肝硬化TPN组比肝硬化对照组升高明显 (P <0 .0 5 )。肝脏NO含量 ,肝硬化对照组比正常组升高明显 (P <0 .0 5 ) ,但肝硬化TPN组比肝硬化对照组降低明显 (P <0 .0 5 )。肝脏 β NADPH黄递酶组化染色 ,正常组为 -,肝硬化对照组为 +,肝硬化TPN组为 ++。结论 NO可能是一种抗肝损伤因子 ,损伤因子存在时 ,表现为肝脏NOS活性增强 ,当肝脏NO含量减少时 ,表现为肝脏损害的加重。Aim To investigate the changes of nitric oxide(NO) and nitric oxide synthase(NOS) activity in cirrhotic rats with TPN and its implication.Methods Wistar rats were divided into 3 groups:normal group( n =8),cirrhotic TPN group( n=7) ,and cirrhotic control group ( n =6).The level of serum and liver NO,liver NOS activity were measured.β NADPH diaphorase staining was observed.Results All animals survived but one rat died of pneumonedeman in TPN group.The level of serum NO and liver NOS activity were significantly higher( P <0.05) in the cirrhotic control group than in the normal group,and also in the cirrhotic TPN group than in the cirrhotic control group.The level of liver NO was significantly higher( P <0.05) in the cirrhotic control group than in the normal group,but significantly lower( P <0.05) in the cirrhotic TPN group than in the cirrhotic control group.The degree of β NADPH diaphorase staining in the liver,normal group was negative,cirrhotic control group was positive(+),cirrhotic TPN group was positive(++).Conclusions NO may be an anti liver injury factor.Liver NOS activity increased when there is injury factor.When the level of liver NO decreases,liver injury will increase.
