皮肤常见恶性肿瘤中端粒酶hTRT的表达

摘　　要：用免疫组化(LSAB)法检测皮肤常见的恶性肿瘤中端粒酶hTRT(humantelomerasereversetranscriptase)的表达特征,探讨hTRT在肿瘤中的可能作用。结果显示,140例肿瘤中hTRT阳性表达率为76.4%107/140,其中鳞状细胞癌75%51/68基底细胞癌72.4%21/29恶性黑素瘤88.2%30/34,Bowen病55.6%5/9均显著高于癌旁相对正常皮肤15.38%4/26。在鳞状细胞癌、基底细胞癌和恶性黑素瘤之间及在同型肿瘤的不同分化、分型中hTRT表达率无显著差异。发现3种hTRT表达模式膜型以鳞状细胞癌为主,阳性细胞多分布在癌巢周围;核型以基底细胞癌为主,阳性细胞多分布在癌巢中央;浆型以恶性黑素瘤为主,阳性细胞呈灶性分布。提示在皮肤常见恶性肿瘤中hTRT表达是个常见事件,肿瘤中hTRT异质性分布与肿瘤类型相关。In order to investigate human telom erase reverse transcriptase(hTRT)gene activity in skin malignant lesions and its pos-sible role in these tumors,the expre ssion of hTRT in 140cases of skin mali gnant lesions by immunohistochemis try technique was in-vestigated.Our results showed that among 140samples of malignant lesio ns 76.4%(107/140)exhibited hTRT.In squamous cell carcinoma,basal cell carcinoma,cu taneous malignant melanoma and Bowe n' s disease,hTRT was exhibited in 75%(51/68),72.4%(21/29),88.2%(30/34)and 55.6%(5/9),respectively,which was significa ntly higher than that in the normal sk in(15.38%,4/26).In this study three types of hTRT dis tribution patterns were found.They were(1)membrance type:major in squamous ce ll carcinoma,positive cells distributed around carcinoma nest;(2)nuclear type:major in basal cell carcinoma,positive cells distributed in the center of nest;and(3)cytoplasmic type:major in malignan t melanoma,positive cells distribu ted in the focal pattern.This suggested that telomerase activation was regarded as a frequent event in skin malignant tumors.The hTRT distribution showed het-erogeneity,which correlated to tum or kinds.
