乙型肝炎病毒S区蛋白肝癌细胞株的稳定表达

摘　　要：构建了乙肝病毒S区不同蛋白的重组真核表达质粒pcLS2、pcLS4,并转染肝癌细胞株HepG2。转染细胞经新霉素筛选后,获得稳定表达S区不同蛋白的细胞株。Westernblotting可分别检测到相对分子质量为39000的大蛋白及24000的主蛋白在pcLS2、pcLS4稳定转染的细胞中表达。免疫组化实验显示这些蛋白均主要呈细胞质分布。Two recombinantspcLS2andpcLS4wererespectivelyconstructedby insertionof preS/Sor S genefragmentinto eukaryoticexpressionvectorpcDNA-3.TherecombinantswerethentransfectedintohepatomacelllineHepG2,andafterthe transformerselectionwithG418,2isolatedcolonieswereobtained.Two largemajorproteinsof HBVwithrelativemolecular massof39000and24000respectivelyweredetectedby Westernblottingin cellextracttransfectedwithpcLS2or pcLS4.Theimmunohistochemicalresultsdemonstratedthatboththeproteinsdistributepredominantlyinthecytoplasm.
