摘  要:目的 了解白细胞介素 - 1β( IL- 1β)和肿瘤坏死因子 - α( TNF- α)在感染性脑水肿 (感脑 )时的变化及其产生的可能机制。方法  99只大鼠分成空白对照组 ( C组 )、生理盐水组 ( NS组 )、百日咳菌液组 ( PB组 )。 NS组和PB组大鼠按注 NS或 PB后断头处死时间又分别分为 3 0 m in、60 min、12 0 min、2 4 0 min4个亚组。EL ISA法测各组大鼠脑组织匀浆中的 IL- 1β和 TNFα蛋白质水平 ,RT- PCR测定脑组织 TNFα m RNA表达水平 ,电泳迁移率改变法( EMSA)检测核因子 - κB( NF- κB)活性。结果 注菌液后 2 4 0 min大鼠脑组织匀浆中的 IL- 1β( 3 84 .1± 3 7.7pg/ g)、TNFα( 768.4± 86.3 pg/ g)的含量明显增高 ,与 C组、NS3 0 min、NS60 min、NS12 0 min、NS2 4 0 min及 PB3 0 m in、PB60 m in和 PB12 0 m in比较差异有非常显著性 ( P<0 .0 1)。 RT- PCR发现 TNFα m RNA表达信号在 PB60 m in组开始增加 ,PB2 4 0 min达高峰。NF- κB活性在 PB60 min组开始增加 ,PB12 0 min和 2 4 0 min达高峰。结论 脑组织中 IL-1β、TNFα含量显著增加与感脑脑损害有关 ,其 IL- 1β、TNFα含量增多与 NF- κB活性增加相关。Objective To understand the relationship between interleukin-1β(IL-1β),tumor necrotic factor-α (TNF-α)and brain damage in brain edema of rats induced by pertussis bacilli. Methods Of 99 rats were divided into 3 groups: sham control group ( C group),normal saline group (NS group),pertussis bacilli group (PB group). The protein levels of IL-1β,TNF-α were assayed with ELISA and TNF-α mRNA expression was examined using RT-PCR in the brain tissue at various time. Activation of nuclear factor-κB(NF-κB) was examined with electrophoretic mobility shift assay (EMSA). Results IL-1β and TNF-α were significantly increased in the PB group at 240 min (384.1±37.7;768.4±86.3pg/g) compared to the C group,NS group at 30min,at 60min,at 120min,and at 240min. The PB group at 240 min were also significantly increased compared to the PB group at 30min,at 60min and at 120min(P<0.01). Expression of TNF-α mRNA began to increased in the PB group at 60 min and reached a peak at 240 min. Activation of NF-κB also began to increased in the PB group at 60 min,reached a peak at 120min and at 240 min. Conclusion IL-1 β and TNF-α may play a role in producing injury in infectious brain edema. The contents of IL-1 β and TNF-α related to activation of NF-κB.
