胃癌中p16、Rb基因蛋白的表达及意义

摘　　要：目的　探讨p16、Rb基因蛋白的表达与胃癌的发生及临床病理参数的关系。方法　采用S P免疫组化方法检测了 80例胃癌及 2 8例癌旁正常胃粘膜组织中p16和Rb蛋白的表达。结果　胃癌中p16、Rb蛋白表达阳性率(分别为 5 8.8%和 5 5 0 % )显著低于癌旁正常胃粘膜 (10 0 % ) (P <0 .0 1) ,Rb蛋白中度阳性或强阳性胃癌p16蛋白中度阳性或强阳性表达率明显低于Rb蛋白阴性或弱阳性胃癌 (P <0 .0 1)。p16蛋白在胃癌中的表达与肿瘤细胞分化程度、Lauren分型和淋巴结转移密切相关。结论　p16、Rb蛋白表达的缺失与胃癌的发生密切相关 ,p16蛋白的表达可用于判断胃癌患者的预后 ,p16和Rb蛋白的表达相互抑制 。Objective To explore the relationship between p16,Rb protein and gastric carcinogenesis and clinicopathogical parameters.Methods S P immunohistochemical methods were used to detect the expression of p16 and Rb protein in 80 cases of gastric cancer and 28 cases of normal gastric mucosa adjacent to them.Results The positive rate of p16 and Rb protein (58.8% and 55.0%,respectively) in gastric cancer were significantly lower than those in normal mucosa adjacent to them(100%)( P <0.01).The moderate and strong positive rate of p16 protein was significantly lower in Rb protein moderate and strong positive tumors than in Rb protein negative and weak positive ones( P <0.01).The expression of p16 protein was significantly associated with differentiation,Lauren type and lymph node metastasis in gastric cancer.Conclusion The loss of p16 and Rb protein expression is associated with gastric carcinogenesis,the expression of p16 protein may act as a prognostic index of the patients and there is a significant inverse relationship between the expression of p16 and Rb protein.
