摘  要:目的 :探讨胃代膀胱术抑制膀胱肿瘤复发的机制。方法 :采用HE染色、荧光染色、DNA电泳、流式细胞术等技术 ,观察了胃液、稀盐酸、阿霉素对膀胱癌细胞系BIU 87细胞凋亡的影响。结果 :发现胃液能诱导体外培养的膀胱癌细胞凋亡 ,细胞形态学观察凋亡细胞数增多 ,DNA电泳出现梯状电泳带 ,流式细胞术可见凋亡峰 ,胃液组细胞凋亡率为 8.0 9% ,与阿霉素组 (4 2 .74% )和无药培养液组 (6 .71% )比较差异有高度显著性 (P <0 .0 1) ,与稀盐酸组 (7.2 6 % )比较差异无显著性 (P >0 .0 5 )。结论 :胃液作为一种诱导膀胱癌细胞系细胞凋亡的因子 ,可能在胃代膀胱术中起到抑制膀胱肿瘤复发的作用。Objectives:To study the mechanism by which gastrocystaplasty inhibits the recurrence of bladder cancer.Methods:The effect of gastric juice,hydrocholoric acid and adriamycin on apoptosis of human bladder cancer cell line BIU 87 were observed by HE,fluorescence,DNA electrophoresis and flow cytometry.Results:Gastric juice induced apoptosis of human cancer bladder cell line in vitro. Apoptotic cells could be seen on light microscopy and fluorescence microscopy.DNA ladder appeared in DNA electrophoresis.Apopotosis peak was observed by flow cytometry.Apoptosis rate of gastric juice group (8.09%) was significantly different from that of adriamycin group(42.74%) and medium group (6.71%),but not significantly different from that of hydrocholoric acid group (7.26%).Conclusion:The results demonstrated that gastric juice could inhibit the recurrence of bladder cancer in gastrocystaplasty as a factor inducing apoptosis of bladder cancer.
