摘 要:目的探讨C-erbB2、C-myc和EGFR多种癌基因共扩增与人体非小细胞肺癌的复发、转移和预后的相关性.方法采用DNA印迹技术(Blot)分别检测156例非小细胞肺癌手术标本及正常肺组织DNA中C-erbB2、Gmyc和EGFR基团扩增现象,并对上述所检测的患者作5年随访追踪以观察多种癌基因扩增与肺癌患者预后的相关性.结果 C-erbB2、Cmyc和EGFR基因中出现双基因以上的共扩增率与肺癌的TNM分期呈正相关,Ⅰ期患者的共扩增率为46%,Ⅱ~Ⅲ期肺癌患者的阳性共扩增率为64%(P>0.05).多种癌基因共扩增呈阴性的肺癌患者生存率高于呈阳性患者,两组间具有明显差异(P=0.001).多种癌基因共扩增率与肺癌的组织学类型、性别和年龄无相关性(P>0.05).结论 C-erbB2、C-myc和EGFR多基因共扩增率与非小细胞肺癌患者的复发、转移和生存期相关.ve To explore the relationship between co-amplification of multiple oncogenes (C-erbB_2、C-myc、EGFR) andprognosis of patients with non-small cell 1ung cancer(NSCLC).Methods The amplification of multiple oncogenes (C-erbB_2、C-myc、EGFR) of l56 patients with NSCLC was determined by use ofDNA Slot blot, and the patients were follow-up for 5 year toobserve the relationship between the co-amplification of multipleoncogenes and prognosis of patients with NSCLC.Results The rateof co-amplification of oncogenes was significantly higher in stage Ⅱ~Ⅲ(63/99, 64%) than in stage Ⅰ(24.5%, 14/57). Survivalanalysis showed that the patients with co-amplification of oncogeneshad worse survival than the patients without co-amplification (P<0.05). No significant difference existed between co-amplification ofoncogenes and sex as well as histological type. Conclusion The co-amplification of C-erbB_2、C-myc and EGFR in NSCLC patients maybe an important molecular events in the carcinogenesis of non-smallcell lung cancer.
