摘 要:目的 研究瘤内单纯注射3 2 P胶体和先注射聚合白蛋白 (MAA) ,再注射3 2 P胶体两种不同给药方法的3 2 P在肿瘤组织和血液及各器官的动态分布 ,探讨MAA减少肝癌组织内3 2 P胶体血行扩散的作用。方法 在Balb/c小鼠右侧胸前皮下接种H2 2 肝癌细胞 ,10天后接种部位长出直径约 1cm的肿瘤。随机将其分为 2组 :第 1组只注射3 2 P胶体 1.85MBq ;第 2组先注射 1× 10 5颗粒MAA ,再注射3 2 P胶体 1.85MBq。注射后的第 30分钟、2 4小时、48小时、第 8天和第 16天时处死小鼠 ,测定肿瘤组织、血液和心、肝、肾、肺、骨骼的放射性。结果 瘤内注射3 2 P胶体时 ,肿瘤内的放射性随时间延长而下降 ,但注射MAA组的放射性下降速度较未注射组明显延缓 ;而其他器官的放射性 ,注射MAA组较未注射组明显减少。结论 与单纯瘤内注射3 2 P胶体相比 ,先在瘤内注入MAA ,再注入3 2 P胶体 ,MAA可以有效阻止3 2 P胶体的全身扩散 ,使3 2 P胶体能够较长时间的滞留在肿瘤内 ,减少了全身分布。Objective To compare deposition and systemic distribution of colloidal 32P in colloidal 32P injection versus injection of macroaggregate albumin (MAA) followed by colloidal 32P,to evaluate the effects of MAA on decreasing diffusion of colloidal 32P from hepatic carcinoma tissue.Methods Balb/c mice were inoculated subcutaneously by H 22 hepatocellular cancer cells in the right fore leg.When the tumors reached 1.0cm in diameter in about 10 days after injection,the mice were divided into two groups randomly.Group A was only injected slowly with 1.85Mq colloidal 32P;B group was injected 1×10 5 particles of MAA followed by 1.85MBq colloidal 32P. The mice of each group were sacrificed at 30min?24h?48h?8d and 16d after injection.The radioactivities of the tumor tissues?blood and heart?liver?spleen?kidney?lung?bone of each animal were measured with a gamma counter.Results Radioactivity in tumor tissuse decreased as time passed.The decreasing speed of radioactivity in tumor tissues in group B was slower than that in group A.The systemic distribution of 32P in group B was lower than that in group A.Conclusion With intratumoral injection of colloidal 32P combined MAA,MAA could retard diffusion of colloidal 32P from tumor tissuse to other organs and deposit colloidal 32P in the tumors for longer time,therefore decrease the distribtuion of colloidal 32P from hepatic carcinoma tissue to other organs.
