摘 要:目的 探讨 6号染色体长臂上与非小细胞肺癌发生相关的微卫星位点。方法 应用多重PCR对 41例非小细胞肺癌中 6号染色体长臂上的 18个微卫星位点进行扩增。PCR产物应用聚丙烯酰胺凝胶电泳分离 ,电泳结果用 Gene Scan TM ,Genotyper TM软件进行分析。结果 各个位点有明显不同的杂合性缺失 (loss of heterozygosity,L OH)频率 ,从 3.85 %到 38.45 %不等 ,总 L OH频率为 5 8.5 % (2 4/ 41)。L OH频率超过 2 0 %的位点共有 8个 ,主要分布在 6 q2 4及 6 q2 7。具体范围为 6 q2 4- 6 q2 5 .3[D6 S16 99(35 % )、D6 S40 9(2 3.33% )、D6 S44 1(33.33% ) ]及 6 q2 6 - 2 7[D6 S15 5 0 (38.45 % )、D6 S2 6 4(2 0 % )、D6 S15 85 (2 5 % )、D6 S44 6 (33.33% )、D6 S2 81(30 .77% ) ]。结论 6 q2 4及 6 q2 7附近可能存在与非小细胞肺癌发生相关的肿瘤抑制基因。Objective: To investigate if there are microsatellite loci in the long arm of chromosome 6 that have close relationship with non-small cell lung cancer. Methods Multiple PCR approach was used to analyze the 18 loci in the long arm of chromosome 6. The PCR products were analyzed in PAGE, and then the electrophoresis maps were analyzed with Gene Scan™ and Genotyper™. Results There were different frequencies of logs of heterozygosity (LOH) in different loci (varying from 3. 85% to 38. 45%). The total frequency of LOH in 41 gastric cancers was 58.5% (24/41). Eight loci with the LOH frequency higher than 20% were mainly located in 2 regions: 6q24 and 6q27. The accurate location is 6q24-6q25. 3 [D6S1699(35%), D6S409(23.33%), D6S441(33.33%)] and 6q26-27 [D6S1550(38.45%), D6S264 (20%), D6S1585(25%), D6S446(33.33%), D6S281 (30.77%)]. Conclusion There may be tumor suppressor genes located in the region of 6q24 and 6q27, which have close relationship with non-small cell lung cancer.
