儿童急性白血病外周血中端粒酶活性的表达及临床意义

摘　　要：目的　探讨端粒酶活性在儿童急性白血病不同时期外周血中的表达及临床意义。方法　采集 5 5例急性白血病患儿和 2 0例对照组儿童的外周血 ,处理后进行端粒重复序列扩增 微孔板杂交分析法。结果　 5 5例不同时期急性白血病患儿 ,初诊 13例中测到端粒酶活性阳性表达 10例(占 76 92 % ) ,复发 9例中测到端粒酶活性阳性表达 7例 (占 77 78% ) ,2 0例对照组及 3 3例缓解期患儿无端粒酶活性表达。初诊时端粒酶活性表达高低与治疗后的缓解情况有一定的相关趋势。 2例初诊时端粒酶活性较高表达者 ,缓解后复发时的端粒酶活性高于初诊时所测。结论　外周血中端粒酶活性的检测 ,对儿童急性白血病的诊断、缓解。Objective To evaluate the expression of telomerase activity in peripheral blood from children with acute leukemia and its clinical significance.Methods Peripheral blood samples were collected from 55 children with acute leukemia in different stages and 20 healthy controls,and evaluated by using TRAP Hyb assay.Results Telomerase activity was positive in 10 of 13 patients (76 92%) at initial evaluation before chemotherapy was employed,and it was negative in 20 controls and in all 33 patients at the period of remission.There was correlation between the level of telomerase activity and the timing of remission after chemotherapy.The telomerase activity was also positive in 7 of 9 patients (77 78%) at relapse,and in two of them the expression of telomerase activity were higher than at initial evaluation.Conclusions Detection of telomerase activity in peripheral blood is of clinical implications to the diagnosis and prognosis of acute leukemia in children.
