摘  要:目的 观察磷脂酶C抑制剂苯甲基磺酰氟 ( phenylmethylsulfonylfluoride ,PMSF)对脑缺血时脑细胞膜游离脂肪酸 (freefattyacid ,FFA)释放的治疗及保护作用。方法 选用雄性SD大鼠局灶性脑缺血模型 ,气 液色谱分析技术观察了脑缺血前后应用PMSF对不同缺血时间点脑细胞膜FFA含量的变化。结果 PMSF主要抑制缺血早期磷脂酰肌醇和缺血后期磷脂酰乙醇胺、磷脂酰胆碱的FFA释放。结论 多不饱和脂肪酸 (C2 0∶4、C2 2∶6 )积聚是反映缺血性脑损伤程度的重要指标 。Purpose: To examine the effect of a phospholipase C inhibitor(phenylmethylsulfonyl fluoride, PMSF) on the neuronal membrane free fatty acid(FFA) during cerebral ischemia in the models of rats. Methods: Before and after operation treated with PMSF, using the model of focal cerebral ischemia in SD rats, the FFA contents in the neuronal membranes were measured in the sham-operated group at 1,5,15,60 and 360 min, examing the content of free fatty acid(FFA) with gas liquid chromatography(GLC) technique. Results: PMSF inhibited the release of FFA primarily from phosphatidylinositol at the earlier stage of cerebral ischemia and from phosphatidylcholine and phosphatidylethanolamine at the latter stage of ischemia. Conclusions: The polyunsaturated fatty acids(C20:4, C22:6) during ischemia is a hallmark of the evolution of ischemic brain damage. The results of the present study supported the existence of certain protective and theraprutic effect of phosphlipase C inhibitor against cerebral ischemia.
