摘 要:目的 :研究转染试剂Lipofectin、DOTAP和FuGENE 6介导反义c myc基因转染人脑胶质瘤细胞U2 5 1的作用。方法 :将可在真核细胞表达人反义c myc基因的质粒pCED4ASCMH分别与转染试剂Lipofectin、DOTAP和FuGENE6结合 ,转染到人脑胶质瘤细胞U2 5 1中 ,转染的第 2 4,48,72和 96小时用MTT比色法检测细胞活性并进行比较。结果 :转染实验组较对照组细胞活性均降低 30 %~ 40 %。结论 :三种转染试剂介导反义c myc基因质粒DNA转染U2 5 1细胞具有抑制细胞增殖、降低细胞活性。Objective:To study the effects of transfection of antisense c myc gene mediated by defferent transfection reagents on human glioma U251. Methods:The plasmid pCEP4ASCMH ascmh expressing human antisense c myc gene in eukaryocyte was transfected to human glioma U251 strain, mediated by liposome Lipofectin, DOTAP and a new transfection reagent FuGENE6, respectively, and the cell activity was assessed by MTT test at 24h, 48h, 72h, and 96h after transfection. Results:It showed that there was a decrease of 30%~40% of the cell activity after transfection mediated by the three reagents when compared with the blank control. Conclusion: The transfection of antisense c myc plasmid DNA mediated by the three transfection reagents inhibited glioma proliferation and glioma activity, thus induced glioma death.
