金地鼠颊囊癌细胞增殖与细胞凋亡的初步研究

摘  要:目的 研究金地鼠颊囊癌细胞增殖与细胞凋亡的改变。方法 建立二甲基苯并蒽 (DMBA)诱导金地鼠颊囊癌模型 ,流式细胞仪 (FCM )检测细胞DNA倍体、S期细胞比例 (SPF)及细胞凋亡 (Apoptosis ,Apop)的变化。结果 颊囊癌的异倍体检出率较正常颊囊黏膜高。原位癌时SPF与Apop较正常颊囊黏膜增高。Ⅰ级鳞癌时 ,SPF较原位癌时不但未增高反而下降 (P <0 .0 5 ) ,而Apop较原位癌下降 (P <0 .0 5 )。Ⅱ级鳞癌时 ,SPF较Ⅰ级鳞癌时增高 (P <0 .0 5 ) ,与原位癌比较无显著性差异 (P >0 .0 5 ) ;而Apop较原位癌时下降 (P <0 .0 5 ) ,与Ⅰ级鳞癌比较无显著性差异 (P >0 .0 5 )。结论 本实验结果提示口腔癌的发生与发展和细胞增殖与细胞凋亡的关系失调有关。它们的进一步研究将有助于了解口腔癌的生物学特性 ,有助于其病理诊断 ,估计预后 。Objective To study the change of cell proliferation and apoptosis of golden hamster cheek pouch carcinoma. Methods DMBA induced golden hamster cheek pouch carcinogenesis model was done .DNA Index (DI),S phase fracture (SPF) and apoptosis (Apop) were detected by flow cytometry (FCM) in cheek pouch carcinoma which included 20 in situ carcinoma (ISC), 21 squamous cell carcinoma (SCC) I grade and 9 Ⅱ grade . Results DNA aneuploidy were found 76% (38/50) in carcinoma, there were significant differences compared with normal (0/10). SPF and Apop in ISC were increased compared with nomal ( P < 0.05 ). Compared with ISC,in SCCⅠSPF were not increased but decreased ( P < 0.05 ), Apop decreased ( P < 0.05 ).In SCCⅡ SPF were increased compared with SCCⅠ( P < 0.05 ), similar with ISC ( P > 0.05 ), but Apop decreaser compared with ISC( P < 0.05 ). Conclusion The results indicate that cheek pouch carainoma happening and development is correlated with dislocation of cell proliferation and apoptosis.The further study of SPF and Apop will be helpful for understand the biological behavior of oral carcinoma, helpful for pathological dialogue and helpful for guide to treatment.
