摘　　要：目的 :构建一种能在肿瘤组织中进行肿瘤特异性表达的逆转录病毒载体 ,增强肿瘤基因治疗的靶向性。方法 :PCR法获得HER2基因的启动子片段并将其克隆至逆转录病毒载体PLXSN中 ,再将突变型绿色荧光蛋白(mtGFP)基因克隆至该启动子下游。结果 :质粒PLHSN经EcoRI和BamHI酶切后可切出 5 10bp和 5 .8kb两条片段 ;质粒PLHGSN经BamHI酶切可切出 5 2 0bp和 6 .7kb两条片段。 结论 :成功构建了携带HER2基因启动子和mtObjective: We aim to construct a retrovirus vector that expresses the downstream gene specifically in tumor cells so as to increase the specificity of gene therapy. Methods: The HER2 promoter fragment was obtained by PCR, then it was cloned into the retrovirus vector P LXSN .The mutant type green fluorescence protein cDNA was subsequently inserted just downstream of this promoter.Results:The plasmid P LHSN can be digested into two fragments with EcoR I and BamH I:510bp and 5.8kb in length respectively; the plasmid P LHGSN can be digested into two fragments with BamH I:520bp and 6.7kb in length respectively. Conclusion:We sucessfully construct a retrovirus vector carrying HER2 gene promoter and mtGFP reporter gene.
