摘  要:目的 :探讨联合应用人白细胞介素 2 (hIL 2 )和人粒细胞巨噬细胞刺激因子 (hGM CSF)基因瘤苗抗肿瘤作用。方法 :经逆转录病毒载体将hIL 2基因及hGM CSF基因导入大鼠Walker瘤细胞WRC2 5 6中 ,制备瘤苗接种荷瘤大鼠。结果 :经PCR SouthernBlot检测证实hIL 2基因及hGM CSF基因已成功导入WRC2 5 6细胞基因组中 ,经生物学活性检测证实转基因细胞具有分泌hIL 2及hGM CSF活性。转基因瘤苗经 6 0GyX线灭活 ,接种荷瘤Walker瘤模型。两种瘤苗确可诱导有效抗肿瘤反应 ,但与单独应用一种瘤苗相比较 ,联合应用无相加或协同作用。结论 :两种瘤苗可诱导有效抗肿瘤反应 。Objective: To investigate the Anti tumour effect of the tumour vaccines engineered by human interleukin 2(hIL 2) and human granulocyte macrophage colony stimulating factor(hGM CSF) genes. Methods: The hIL 2 and hGM CSF genes were transferred into the Walker tumour cell vaccinesbby virtue of retrovirus. The gene transferred vaccines were inoculated into the Walker tumour model.Results:Gene transfers were successfully confirmed by PCR Southern Blot. The biological activities of hIL 2 and hGM CSF by the gene transferred vaccine cells were showed with MTT assay. After in Vitro irradiation with 60Gy X ray, the gene transferred vaccines were inoculated into the Walker tumour model. Conclusion:The effective anti tumour response was induced by the vaccines. The plus or coordinate anti tumour effect by synchronous inoculation with two kinds of the vaccines was not observed.
