摘 要:本实验旨在研究尾加压素Ⅱ (uⅡ )在慢性低氧性肺动脉高压及肺血管结构重构形成中的变化。应用免疫组化技术对慢性低氧性肺动脉高压大鼠不同节段肺内动脉uⅡ蛋白表达进行定位及半定量分析。结果显示 :低氧 1周和 2周组大鼠肺动脉平均压 (mPAP)、右心室肥厚指标R (L +S)、肺动脉相对中膜面积 (RMA)和相对中膜厚度(RMT)均明显高于对照组 (P <0 0 0 1 )。低氧 2周组大鼠各节段肺内动脉内皮细胞中uⅡ表达较对照组分别下降2 7 75 %、32 50 %、39 63 % (P均 <0 0 1 )。相关分析显示 :与呼吸性细支气管伴行的肺动脉内皮细胞uⅡ表达与mPAP呈显著负相关 ;各节段肺动脉内皮细胞uⅡ表达分别与各级肺动脉RMA、RMT呈明显负相关 (P均 <0 0 5)。研究表明 :低氧性肺动脉高压及肺血管结构重构形成过程中 ,肺内动脉内皮细胞uⅡ表达呈现明显下调 ;To investigate the alteration of urotensin Ⅱ(uⅡ)in the intrapulmonary arteries of rats with hypoxic pulmonary hypertension and pulmonary vascular structural remodeling, expression and localization of uⅡ in the different segments of intrapulmonary arteries of hypoxic pulmonary hypertension rats were studied by immunohistochemistry The mean pulmonary artery pressure(mPAP), right ventricular hypertrophy index, RMA and RMT were significantly increased in 1 and 2 weeks hypoxic group compared with those in normal controls( P <0 001) After 2 weeks of hypoxia, uⅡ staining presented in the endothelial cells of intrapulmonary arteries was significantly decreased comparing with normal controls( P <0 01) uⅡ staining was negatively correlated with RMA and RMT of pulmonary arteries at each segment of intrapulmonary arteries( P <0 05) It was concluded that uⅡ staining presented in the endothelial cells of intrapulmonary arteries was decreased significantly with hypoxic pulmonary hypertension and pulmonary vascular structural remodeling, and endogenous uⅡ may be of great importance in the pathogenesis of hypoxic pulmonary hypertension
