摘 要:为研究骨组织主要成分对其 Masson三色染色结果的影响 ,取大鼠股骨上端及尾部中段标本 ,经 Bouin固定液固定 ,甲酸脱钙后 ,石蜡切片 ,对照片染 Masson三色及 Safranin- o染色 ,实验片经过 0 .5 %透明质酸酶消化 ,2 mol·L- 1氯化钙溶液抽提粘多糖后 ,染色同对照片。另取人脐带标本 ,经 Bouin液固定 ,石蜡切片 ,实验片经透明质酸酶消化后 ,做 Safranin- o染色 ,对照片不经透明质酸消化 ,其余处理及染色同实验片。结果示 Safranin- o对组织中粘多糖染鲜红色 ,经透明质酸酶消化及氯化钙溶液抽提 ,其着染减退或消失。 Masson三色染色 ,软骨胶原呈绿色 ,真皮、骨及钙化软骨中胶原呈红 -绿相间 ,骨质越成熟 ,红染越多。经透明质酸酶消化及氯化钙溶液处理染色无改变。表明骨组织中胶原的成熟程度影响 Masson三色的染色反应 ,胶原越成熟红染越多 ,胶原类型 ,粘多糖及钙化成分不影响骨组织对To study the influence of the main components of bone tissues on the Masson's staining results. The specimens were harvested from rat upper femoral end and the middle segment of the tail, fixed by Bouin's solution, decalcified by formic acid and cut by paraffin section methods. The control sections were stained with Masson's and safranin O staining methods. The experimental sections were digested by hyaluronidase and extracted by 2mol/L calcium chloride solution, then stained with the same methods. In addition, human umbilical cord specimens were taken, fixed by Bouin's solution, cut by paraffin section methods. The experimental sections were digested by hyaluronidase and stained by safranin O staining method and the control sections not digested by hyaluronidase were stained with the same method as that for the experimental sections. Results were that the mucopolysaccharide in the bone tissues was dyed in red by safrnin O staining and faded or disappeared after digested by hyaluromidase and extracted by calcium chloride solution; the collagen in the dermis, bone and calcified cartilage was dyed in red alternating with green by Masson's staining method. The more the bone was matured, the more the red dyeing was. The dyeing had no change after hyaluronidase digestion and calcium chloride solution treatment. The study approved that the maturation degree of collagen in the bone tissues affects Masson's staining reaction. The more the collagen is matured, the greater the red dyeing area is. The collagen type, mucopolysaccharide and calcium have no influence on the reaction of the bone tissues to Masson's staining.
