摘 要:目的 :探讨端粒酶活性在儿童急性白血病中的表达及临床意义。方法 :采用端粒重复系列扩增法(TRAP)检测了 11例儿童急性白血病化疗前、完全缓解后骨髓单个核细胞 (MNCs)端粒酶活性水平 ,对部分病例进行追踪观察 ,并与 10例正常儿童骨髓 MNCs端粒酶活性相比较。结果 :正常骨髓 MNCs具低水平的端粒酶活性 ,相对端粒酶活性范围 2 .6 %~ 19.1% ;急性白血病患儿化疗前相对端粒酶活性明显增高 40 .4%~ 98% ,完全缓解后其相对端粒酶活性为 2 1.7%~ 6 0 .1% ,随缓解时间的延长 ,端粒酶活性逐渐降低。结论 :端粒酶激活在儿童急性白血病的发生中可能具有重要作用 ,动态观察端粒酶活性水平 ,对估测病情。Objective:To explore the expression and clinical significance of telomerase activity in childhood acute leukemia.Methods:By means of a PCR based telomeric repeat amplification protocol(TRAP)assay.It is detected telomerase activity in bone marrow mononuclear cells(MNCs)from 11 pediatric acute leukemia at diagnosis and after complete remission,and 4 cases to be trended.For comparative analysis,MNCs from 10 normal children were detected.Results:Normal MNCs from normal children expressed a very low telomerase activity,with the relative telomerase activity in the range of 2.6%~19.1%.High levels of telomerase activity was detected in MNCs from all acute leukemia patients at diagnosis,with the relative telomerase activity in the range of 40.1%~98%.After induction chemotherapy and complete remission,telomerase activity decreased to low levels of 21.7%~60.1%.With prolongation of complete remission,telomerase activity further decreased gradually.Conclusion:Telomerase activation may play a important role in leukemogenesis.To detect regularly telomerase activity in MNCs from acute leukemia patients may be of actual significance for evaluating patient's conditions and monitoring minimal residual disease.
