转化生长因子β1对口腔鳞癌细胞系基质金属蛋白酶-2和-9活性的影响

摘　　要：目的　探讨TGF - β1和口腔鳞癌侵袭转移的关系。 方法　采用蛋白酶谱分析法测量不同浓度的TGF - β1作用于口腔鳞癌TSCCa细胞系及颈淋巴转移癌GNM细胞系后MMP - 2和MMP - 9的活性变化。 结果不同浓度的TGF - β1作用于TSCCa 12小时后 ,MMP - 2和MMP - 9的活性与对照组相比无明显差异 ( P >0 .0 5 ) ;而不同浓度的TGF - β1作用于GNM细胞 12小时后 ,MMP - 2和MMP - 9的活性与对照组相比 ,有显著性差异 (P <0 .0 5～ 0 .0 1) ,MMP - 2和MMP - 9活性与TGF - β1有剂量依赖关系。 结论　TGF - β1可能与口腔鳞癌侵袭转移密切相关。Objective To detect the role of transforming growth factor β1(TGF-β1)on the invasiveness and metastasis of human oral squamous cell carcioma.Methods Using Zomography to measure the activity of matrix metalloproteinase-2 and -9 in oral squamous cell carcinoma cell lines TSCCa and metastasis carcinoma cell lines GNM treated by different levels of TGF-β1.Results TGF-β1 had no effect on the activity of MMP-2 and MMP-9 in TSCCa cell lines,While TGF-β1 had increased the activity of MMP-2 and MMP-9 in GNM cell lines.Moreover,the activity of MMP-2 and MMP-9 had more dependent on TGF-β1. Conclusion TGF-β1 may be a factor associated with invasive and metastasis potential of oral squamous cell carcinoma.
