摘  要:目的观察培养人视网膜色素上皮细胞(RPE细胞)核因子κB(Nucleartranscriptionfactor-κBNF-κB)的表达,及地塞米松对其表达的影响。方法培养人RPE细胞接种于24孔板,随机分为阴性对照组、脂多糖(LipolysaccharideLPS)刺激组和地塞米松及LPS作用组,做免疫组化ABC法染色,显微镜下观察。结果对照组NF-κB在培养RPE细胞胞浆中表达,LPS刺激后转入胞核表达,地塞米松作用后,再以LPS刺激,RPE细胞核及细胞浆内NF-κB的表达减弱,胞核中表达水平减弱明显,且减弱的程度与地塞米松浓度有关。结论LPS能够激活RPE细胞的NF-κB使其向核内转入,一定浓度的地塞米松对RPE细胞的NF-κB表达及向胞核转入有抑制作用。Aim To observe the expression of nuclear factor-κB in human retinal epithelial cells (HRPE)and effects of desamethasone on its expression. Methods Cultured HRPE were implanted in 24 well culture plates with a cover slide and divided into normal control groups, the Lipolysaccharide (LPS) stimulated groups and groups stimulated with LPS and desamethasone. Then the NF-κB in the cells were immunohistochemically stained with ABC technique. Results NF-κB was localized in the cytoplasm of All cells in normal control groups. After stimulated with LPS, the nuclear was markedly stained. When cultured with desamethasone for 40 min and then stimulated with LPS, the staining of nuclear and cytoplasm was attenuated and nuclear staining's attenustion was marked. The degree of attenuating is correlated with the concentration of desamethasone. Conclusion Nuclear factor(NF-κB)exist in RPE cultured in vitro, which can be activated by LPS, the activating of NF-κB can be blocked by desamethasone.
