摘 要:目的研究Doxycycline抑制雄性激素非依赖型前列腺癌细胞PC-3细胞金属蛋白酶蛋白酶的表达及其与体外侵袭转移能力的关系。方法以免疫组织化学方法和transwell小室法研究不同浓度的Doxycycline对PC-3中金属蛋白酶的表达的影响及对体外侵袭转移能力的影响。结果免疫组织化学方法检测结果表明Doxycycline可以抑制PC-3细胞对金属蛋白酶蛋白的表达,transwell小室法结果显示Doxycycline可以抑制PC-3的侵袭转移能力,且两者均具有浓度依赖性。结论Doxycycline可以抑制PC-3的侵袭及转移,与其抑制金属蛋白酶的表达有关。为Doxycycline治疗前列腺癌提供了实验依据。Aim To study the relationship between doxycycline inhibited expression of MMP-2 in androgen-independent prostatic carcinoma cell line PC-3 and doxycycline inhibited invasion and metastasis of PC-3 in vitro. Method Immunohistochemistry and transwell chamber were applied to investigate the expression of MMP-2 in different concentration of doxycycline- treated PC-3 cells and the invasive ability of different concentration of doxycycline- treated PC-3 cell. Result The positive cell of MMP-2 in PC-3 cells was decreased and the cell move through the membrane were decreased with the increasing of exposed concentration of doxycycline. Conclusion Doxycycline can inhibit the invasion and metastasis of PC-3 cells, which is related to the inhibition of MMP-2 in PC-3 cell.
