摘 要:目的 了解胎儿发育期间肾上腺的抗氧化功能及其特征。方法 差速离心法制备胎组织亚细胞组分 ,测定多种抗氧化酶活性 ,并进行相关性分析。结果 肾上腺细胞胞浆内谷胱甘肽S 转移酶 (GST)α、π和 μ亚型活性分别是肝细胞浆的 0 .5倍、4 .4倍和 8.3倍 ,胎肾上腺和肝脏胞浆πGST活性皆随胎龄递减 (r =- 0 .5 79;r =- 0 .6 32 )。微粒体和线粒体中也可检测到πGST和πGST。肾上腺中超氧化物歧化酶、谷胱甘肽过氧化物酶和谷胱甘肽还原酶活性分别为肝脏的 2 .1倍、1.8倍和 5 .0倍 ,而过氧化氢酶活性则明显低于肝脏。电镜下显示 ,8月龄胎肾上腺细胞核周粗面内质网及游离核糖体明显增多 ,可见多个溶酶体 ,次级溶酶体和吞噬小体。结论 胎儿发育时期 ,肾上腺已具有较完善的抗氧化酶系统 。AIM To understand the capacity of adrenal gland to treat various active oxygenic free radicals during the fetal development. METHODS Subcellular fractions of fetal ad renal and liver were prepared by differential cen trifugation. The activities of various antioxidative enzymes were measured. RESULTS The mean values(mmol·min -1 ·g -1 protein) of cytosol αGST (2.20±0.61), πGST (115±62) and μGST (50±32) with cumene peroxide, ethacrynic acid and bromsulfophthalein as the selective substrates in adrenals were 0.5 fold, 4.4 fold and 8.3 fold (P<0.01), respectively, of those in liver. The activities of adrenal and liver cytosol πGST were negatively correlated with fetal ages. αGST And πGST also were detected in the microsomes and mitochondria of fetal adrenals. The cytosol activities of superoxide dismutase, glutathione peroxidase and reductase in adrenals were almost 2.1 fold, 1.8 fold and 5.0 fold higher (P<0.01), however, catalase was lower (19%), than that in liver. CONCLUSION The tissue difference of substrate selectivity occurred between adrenal and liver with more perfect antioxidative functions during the development, and fetal adrenals possess greater antioxidative capacity, which was not weaker than that of fetal liver.
