胃癌组织中IL-8基因的表达及临床意义

摘　　要：目的　探讨趋化因子IL 8与胃癌的浸润深度、转移能力和临床分期的关系。方法　采用逆转录聚合酶链反应 (RT PCR)半定量的方法 ,检测IL 8mRNA在胃癌组织及其相应的正常胃粘膜组织和转移淋巴结中的表达。结果　 2 9例胃癌组织和2 0例转移淋巴结中 ,分别有 2 6例 (89.7% )和 17例 (85 .0 % )IL 8mRNA的表达较相应正常胃粘膜组织强 ;IL 8在胃癌组织中的表达与胃癌TNM分期有关 (P <0 .0 1) ,与肿瘤浸润深度呈正相关 (γ =0 .5 6,P <0 .0 1) ,IL 8基因在转移淋巴结中的表达与肿瘤浸润深度也呈正相关 (γ =0 .5 2 ,P <0 .0 1)。结论　IL 8mRNA的高表达与胃癌的浸润深度、临床分期相关 。Objective To study the overexpression of chemokine interleukin 8(IL 8) mRNA in human primary gastric carcinoma and metastatic lymph node and the relationship between its epression and the infiltration and stage of the cancer.Methods Reverse transcription polymerase chain reaction(RT PCR) was used to detect the expression of IL 8 mRNA in cancer tissue,corresponding normal mucosa and metastatic lymph node of 29 patients.Results The expression levels of IL 8 mRNA in 89.7%(26/29) cancer tissue was much greater than in normal mucosa cases and the expression in 85.0%(17/20) metastatic lymph node was greater than in corresponding normal mucosa,but it was no greater in negative lymph node.The expression level of IL 8 mRNA was positively correlated to clinical pathologic classification and invasive depth.Conclusion These findings suggest that the overexpression of chemokine IL 8 in gastric cancer was correlated to tumor invasive and the TNM classification,in addition,IL 8 mRNA expression was also correlated to tumor invasive depth,so IL 8 may prove to be a new useful marker for evaluating biological aggressiveness and lymph node metastasis of gastric carcinoma.
