摘  要:目的 研究rasp2 1蛋白在bcr ablp2 10蛋白抗凋亡信号传导通路中的作用。 方法 应用逆转录病毒载体pLXSN将反义N ras1cDNA转导到K 5 62细胞系中后 ,通过足叶乙甙对细胞进行诱导凋亡。结果 转导反义N ras1cDNA的K 5 62细胞内N rasp2 1表达下降。反义N ras转导细胞较对照细胞K 5 62易发生凋亡 ,对药物的敏感性显著增高。 结论 rasp2 1蛋白是bcr ablp2 10蛋白介导抗凋亡信号的重要的下游蛋白 ,阻断该蛋白的功能有可能成为治疗人类慢性髓性白血病的Objective In order to investigate the role of ras p21 playing in transduction of the anti apoptotic signals of bcr abl p210.Methods The antisense expression vector antisense N ras1 pLXSN was constructed by N ras1 cDNA and retrotranscription virus vector pLXSN,and was transferred into K562 cell line by means of calcium phosphate co precipitation.The drug etoposide was used to induce cell apoptosis.Results The expression of N ras1 protein was reduced significantly in antisense N ras1 transduced K562 cell.The antisense N ras1 transduced K562 cell was shown to be more susceptible to apoptosis and be more sensitive to the drug than the control K562 cell.Conclusion The ras p21 is one of the main downstream protein in mediating the anti apoptotic signals of bcr abl p210,and blocking ras p21,may become a selective point of action for the treatment of human chronic myeloid leukemia.
