摘 要:目的 探讨非肥胖性糖尿病 (None obesediabeticNOD)小鼠的肝组织学改变及其与空腹血糖的关系。方法 2 6只NOD小鼠 ,低脂低蛋白饮食饲养于屏障系统内。观察至发生显性糖尿病或 9月龄时处死。分析肝细胞脂肪变性与空腹血糖的关系。结果 实验结束时 ,11只小鼠空腹血糖水平始终正常 ,15只 ( 5 7.7%)发生糖尿病。糖尿病组空腹血糖、糖化血红蛋白较血糖正常组显著升高 ,两组小鼠体重无显著差异。苏木精 伊红染色显示肝组织学改变基本正常者 7例 ( 2 6 .9%) ,其余小鼠均有泡沫样和 (或 )大泡性肝细胞脂肪变性 ,8例 ( 30 .8%)达到脂肪肝的诊断标准。相关分析显示 ,随着NOD小鼠血糖水平升高 ,肝脂肪变性程度呈加剧趋势 ;糖尿病组肝脂肪变程度显著重于血糖正常组 (秩和检验H =14 .0 84,P <0 .0 0 1)。脂肪肝组 (n =8)血糖和糖化血红蛋白显著高于非脂肪肝组 (n =17) ,分别为 2 6 .10±7.12对 12 .31± 10 .0 0mmol/L(t =3.0 5 7,P <0 .0 1)和 6 .90± 1.6 6对 3.91± 2 .11mmol/L(t =3.5 47,P <0 .0 1)。结论 NOD小鼠是研究 1型糖尿病与脂肪肝相关性的良好模型 ,空腹血糖水平与NOD小鼠肝脂肪变性程度密切相关。Objective To explore the relationship between blood glucose with liver steatosis in non-obese diabetic (NOD) mice. Methods NOD mice(n=26) lived in a isolated system fed with low fat and low protein diet, and were not sacrificed until to suffered from diabetes or 9 months old. Hepatic histopathology and their relationship with fasting blood glucose concentration in NOD mice were studied.Results At the end of the experiment, there were 11 NOD mice with normal fasting blood glucose and 15 NOD mice with diabetes (57.7%). Both blood glucose concentration and components of adult glycated hemoglobin (HbA1C%) in diabetes group were significantly higher than those in normal blood glucose group, and there is no difference of body mass between two groups. Hematoxylin and eosin stain of formalin-fixed, paraffin-embedded liver biopsy sections, revealed that 7 samples without histologic abnormalities (26.9%), and other samples had microvesicular and/or macrovesicular steatosis, and there were 8 samples with fatty liver according to histologic diagnosis criteria( 30.8%). The degree of steatosis in NOD mice were severe with increased blood glucose level. The degree of steatosis in diabetes group was more severe than that in normal blood glucose group (H=14.084, P<0.001). Furthermore, both blood glucose and HbA1C(%) in fatty liver group(n=8) were higher than those in non-fatty liver group (n=17), 26.10±7.12 vs 12.31±10.00mmol/L (t=3.057, P< 0.01) and 6.90±1.66 vs 3.91±2.11mmol/L (t=3.547, P< 0.01), respectively. Conclusion NOD mice is a good model for study the association of type 1 diabetes with fatty liver. There is a positive correlation between fasting blood glucose and liver steatosis in NOD mice.
