摘 要:的 :探讨去除雄激素对人前列腺癌 ( PCa)细胞中凋亡抑制蛋白 Bcl-2和 Bcl-XL 蛋白表达的影响。方法 :采用雄激素依赖 PCa细胞株 L NCa P为研究对象 ,雄激素非依赖 PCa细胞株 PC-3为对照 ;以 Western印迹法检测去除雄激素后 Bcl-2和 Bcl-XL 蛋白的表达。 结果 :在 L NCa P中去除雄激素 12 h后 Bcl-2蛋白的表达开始升高 ,到 2 4h最明显 ,而去除雄激素 10h后 Bcl-XL 蛋白表达开始降低 ,在 2 4h达最低。而在 PC-3中去除雄激素前后未见 Bcl-2和 Bcl-XL 蛋白表达改变。 结论 :去除雄激素可促 PCa细胞中 Bcl-XL 蛋白表达降低 ,可能是手术或药物去势治疗前列腺癌的机制之一 ,Bcl-2蛋白的表达升高可能和后期 PCa雄激素非依赖性生长有关。Objective: To study the effect of withdrawal of androgen on expression of anti apoptotic protein Bcl 2 and Bcl X L in human prostate carcinoma cells. Methods: Expression of Bcl 2 and Bcl X L in human androgen dependent prostate carcinoma cell line LNCaP was measured by using Western blot analysis after withdrawal of androgen. Human androgen independent prostate carcinoma cell line PC 3 was also examined as control. Results: Down regulation of Bcl X L was detected in LNCaP cell line 10 h after withdrawal of androgen and the lowest expression level of Bcl X L was found after 24 h. Up regulation of Bcl 2 was found in LNCaP cell line after 12 h of withdrawal of androgen, and reached the highest after 24 h. No change of expression of either Bcl 2 or Bcl X L was found in PC 3 cell line after withdrawal of androgen. Conclusion: Withdrawal of androgen can down regulate the expression of Bcl X L, which may be one of the mechanisms by which prostate carcinoma can be controlled after androgen ablation by either operative or medicine methods. Expression increase of Bcl 2 may be one of the mechanisms by which androgen independent PCa is developing.
