摘  要:目的 建立特异的大鼠T细胞系 ,为T细胞疫苗的研制打下基础。方法 采用足垫皮内注射豚鼠全脊髓匀浆 (GPSCH) 福氏完全佐剂 (CFA)混合乳剂和百日咳疫苗 ,诱导Wistar大鼠EAE模型。腹股沟淋巴结细胞经豚鼠髓鞘碱性蛋白 (GPMBP)反复刺激 ,3H TdR掺入法筛选针对GPMBP特异增殖的T细胞。FACS检测T细胞表型。RT PCR测定T细胞的细胞因子格局。结果 一次性足垫皮内注射GPSCH CFA混合乳剂可诱导Wistar大鼠实验性自身免疫性脑脊髓炎 (EAE)模型 ,并证实了同时注射百日咳疫苗能提高发病率和发病程度。EAE大鼠脑、脊髓组织HE染色见大量淋巴细胞浸润 ,在血管周围形成淋巴鞘 ,但未见脱髓鞘改变。T细胞对GPMBP呈特异性增殖 ,为CD4 +表型。RT PCR结果显示T细胞IL 2和IFN γmRNA表达水平较高 ,TNF α亦低水平表达。结论 Wistar大鼠可用于制备EAE动物模型。GPMBP特异性T细胞系为CD4 +表型 ,呈TH1类细胞因子格局。Objective As a technical plateform of T cell vaccine preparation, we made up a MBP specific rat T cell lines. Methods Wistar rats develop EAE model was established by immunization of guinea pig spinal cord homogenate(GPSCH) in CFA and pertussis vaccine(PV). LNC was stimulated by GPMBP in every two weeks, and selected by 3H thymidine incorporation. The surface phenotype of the MBP line was determined by FACS. The cytokines mRNA expression was assayed by RT PCR. Results A single injection with GPSCH in CFA and PV induced EAE in Wistar rats. GPMBP specific T cell lines were produced. Their phenotype were CD4 +. The mRNA expression for IL 2 and IFN γ was higher than that for TNF α. Conclusion EAE could be induced in Wistar rats. The surface phenotype of the T lymphocyte lines was CD4 +, and showed a T H1 cytokine profile.
