摘 要:为了探讨大鼠局灶性脑缺血再灌注缺血半暗带β淀粉样前蛋白 (APP)转录水平与缺血时间及梗死体积的相互关系 ,用插线法建立大鼠局灶性脑缺血再灌注模型 ,剥取缺血半暗带皮质组织 ,采用半定量逆转录 聚合酶链式反应 (RT PCR) ,测定永久性缺血 4 8h和不同缺血时间再灌注 4 8h后 ,APPmRNA水平的变化。结果显示 ,梗死体积随再灌注前缺血时间的延长而增大 ,皮质半暗带缺血 30min再灌注 4 8hAPPmRNA表达升高 ;缺血 6 0min和缺血 12 0min再灌注 4 8hAPPmRNA升高明显 ;缺血 180min再灌注 4 8h和永久性缺血 4 8hAPPmRNA达到高峰。提示缺血半暗带APPmRNA的表达随再灌注前缺血时间延长而增加并与梗死体积有一定的相关性 ,早期再灌注可减少其表达。 ((GFDA1) )In order to study the relationship between amyloid precursor protein (APP) transcription level in rats'ischemic penumbra and infarct size, focal ischemia models of middle cerebral artery occlusion (MCAO) were established by inserting nylon surgical thread through the internal carotid artery (ICA) into the anterior cerebral artery (ACA). After different time of left MCA occlusion, the thread was removed to allow reperfusion.Brain samples were harvested from ischemic penumbra. The expression of APP mRNA was assessed by RT PCR. It was found that cerebral infarct size increased according to ischemic time before reperfusion. Following 48 h reperfusion APPmRNA of the penumbra regions was higher than their normal level after 30 min' and 60 min' ischemia, and significently enhanced after 90 min' ischemia. It reached the top in 180 min' ischemia and permanent ischemia. It suggested that there might be a correlation between APPmRNA level and infarct size, and early reperfusion could reduce APPmRNA expression.
