摘 要:目的 :初步探讨机械压力对成骨样细胞增殖活性及功能状态的影响 ,为研究正畸矫治力对牙周及骨组织的生长改建的机理提供理论依据。方法 :对体外培养的成骨样细胞在受到顶—底轴向压力 (2 2 5×g)后 6、12、18、2 4、30、36h ,利用流式细胞术及MTT比色法 ,检测细胞增殖动力学的变化及线粒体琥珀酸脱氢酶的代谢活性。结果 :加力组较对照组各期细胞所占百分比及细胞功能状态都有显著性差异 (P <0 0 1) ,其中以加力后 12h、S期细胞的增加及功能状态的降低最为明显 ,而增殖指数增加的最大值出现在加力后 18h ,加力组与对照组各项指标在 2 4h后趋于一致 ;前向散射光参数FSC两组间无显著性差异。结论 :顶—底轴向压力 (2 2 5×g)可显著影响成骨样细胞的增殖活性和功能状态 ,并随时间的推移逐渐恢复 。Objective:The aim of this study was to investigate the effects of the mechanical stress on the proliferation and function of osteoblast_like cells (UMR106) of rats, and further to explore the mechanism of remodeling of periodontium from an angle of mechanics. Methods:The flow cytometry and MTT method was used to examine the number of osteoblasts_like cells and their proliferation and metabolic enzymic activity 6 h, 12 h, 18 h, 24 h, 30 h, and 36 h after the top_bottom axial stress (225×g) was exerted on the UMR106 in vitro.Results:The difference of cell phase fraction and function between the experimental group and the control group were of great significance (P<0 01). The maximum SPF increasing and functional decreasing appeared 12 h after the appliance of the stress, and all indexes were no significantly different between the two groups 24 h after the stress was supplied. Forward light scatter had no remarkable alteration.Conclusion:The top_bottom axial stress (225×g) could inevitably influence the proliferation and function of osteoblast_like cells without pathological damage occurred.
