摘 要:目的 研究川芎嗪 (TMP)对人多形核白细胞 (PMN)呼吸爆发期间氧消耗和活性氧自由基的影响。方法 用PMA刺激PMN呼吸爆发产生的活性氧自由基 ,黄嘌呤 /黄嘌呤氧化酶体系产生的超氧阴离子 (O2 -)和Fenton反应产生的羟自由基 (OH-)为模型。用电子自旋共振自旋捕集技术 ,自旋探针氧测定法和化学发光法研究TMP对活性氧自由基O2 -和OH-的清除作用及对PMN呼吸爆发时氧消耗的影响。结果 TMP(0 .18mol·L-1)对两种水溶液体系产生的O2 -和OH-的清除率分别达 10 0 %和 44 % ,也能显著清除细胞体系产生的活性氧自由基 ,抑制它的化学发光过程 ,但对PMN呼吸爆发时的氧消耗无明显影响。结论 TMP不影响PMN呼吸爆发时的氧化代谢 ,但能通过清除氧自由基防止激活PMN对组织的损伤。Objective To investigate the influence of ligustrazine (tetramethylpyrazine, TMP) on oxygen consumption during the respiratory burst of human polymorphonuclear leukocytes (PMN) stimulated with PMA and active oxygen free radicals.Methods Influence of TMP on superoxide anion (O 2 -),hydroxyl radicals (OH -) and oxygen consumption were studied by using ESR spin trapping, spin probe oxymetry and luminol dependent chemiluminescence (CL). The active oxygen free radicals were generated from respiratory burst of PMN stimulated with PMA, O 2 - generated from xanthine/xanthine oxidase (Xan/XO) system and OH - generated from Fenton reaction.Results The scavenging rate of O 2 - by TMP ( 0.18 mol·L -1 ) was 100% in Xan/XO system, and that of OH - was 44% in Fenton reaction. TMP ( 0.18 mol·L -1 ) had no influence on oxygen consumption during the respiratory burst of PMN, but had remarkable inhibition on CL response generated by PMN, and had scavenging effect on active oxygen free radicals generated by PMN.Conclusion TMP has no inhibitory effect on oxygen metabolic function of PMN, but can prevent tissue injury caused by activated PMN through scavenging oxygen radicals.
