摘  要:目的 研究甲基硝基亚硝基胍 (MNNG)对日本血吸虫成虫培养细胞碱性磷酸酶 (AKP)和酸性磷酸酶 (ACP)活性的影响。方法 将虫龄 2 6 d的日本血吸虫成虫细胞接种于小盖玻片上 ,在RPMI- 16 40含 2 0 %小牛血清附加常量抗生素的常规培养基中培养第 7天时 ,随机分为实验组和对照组 ,实验组用含浓度为 3μg/ ml MNNG的常规培养基处理 48h,对照组用不含 MNNG的常规培养基处理同样时间 ,随后换用常规培养基培养 3周 ,当再换用含 5 %小牛血清的培养基培养 1周时 ,分别用 Gomori钙钴法和 Gom ori硫化铅法对两组培养细胞进行 AKP和 ACP细胞化学染色 ,用Olym pus BH- 12显微镜观察并拍照 ,用 HPIAS- 2 0 0 0图像分析仪测量其含量 ,并作统计分析。结果 实验组培养细胞的 AKP、ACP活性明显高于对照组培养细胞的活性 (P<0 .0 1)。结论  MNNG能增强日本血吸虫成虫培养细胞 AKP、ACP的活力 ,对培养细胞有促生长或 /和诱导其转化的作用。Objective To study the influence of MNNG on the activities of alkaline phosphatase (AKP) and acidic phosphatase (ACP) in the cultured cells from adult Schistosoma japonicum. Methods The cells from 26d adult Schistosoma japonicum were inoculated on the small glass coverslips. After culture for 7 days in RPMI 1640 containing 20% calf serum and a moderate amount of antibiotics, the cells were divided into test and control groups. The cells in the test group were treated by MNNG with concentration of 3 μg/ml for 48 hours; the cells in the control group were treated as same as the test group except MNNG in the media. Then the cells were cultured in normal media for 3 weeks. When they were transferred into the media of RPMI 1640 with 5% calf serum and a moderate amount of antibiotics, and cultured for another week, the cells in two groups were stained to show the activities of AKP and ACP by the methods of Gomori Caco and Gomori Pbs respectively. The contents were measured by using a photometer image analytical instrument (HPIA 2000) and analyzed by statistical method. Results The activities of AKP and ACP in the cultured cells of test group were significantly higher than that of control group (P <0 01). Conclusion MNNG could increase the activities of AKP and ACP in the cultured cells and gave an impetus to the cell growth or/and transformation.
