摘 要:本研究对我国两株登革2型病毒D2-43株、D2-04株的基因组进 行 了全序列测定,在此基础上对这两株引起不同临床症状及鼠神经毒力的登革病毒的基因组序 列进行了比较分析,结果表明D2-43株与D2-04株基因组全长约为10 723nt,核苷酸序列的同源性为95.1%,氨基酸序列的同源性为97.6%, 不存在特别的高变区。这两株序列中共有83个核苷酸的变化导致了氨基酸的变化,其中21个 差异氨基酸可引起所在位点电荷或极性的变化,位于登革病毒粒子表面的E糖蛋白第126位氨 基酸由Glu (D2-04株)→Lys(D2-43株)的变化对其抗原性有影响,可能引起了病毒对鼠神 经 毒力的改变。对结构糖蛋白E基因的聚类分析表明D2-43株与新几内亚株、台湾87株及菲律 宾 83株亲缘关系较近,D2-04株与牙买加株及巴西90年分离株的亲缘关系较近,表明我国存 在不同起源的登革2型病毒感染。The complete genomic sequences of D2-43 and D2-04 strai ns isolated in China were determined in this study. Further we compared the full n ucleotide sequences of D2-43 and D2-04 strains, the results showed that their ge nomic length was 10723 nucleotides, nucleotide sequence homology was 95.1% and t he nucleotide differences were scattered throughout the entire sequences of the genomes. The amino acid sequence homology was 97.6% and this gave rise to 83 am ino acid substitutions. There were 21 amino acid substitutions that confer chang es in charge or side chain polarity. Computer analysis of the amino acid changes in structural proteins revealed a change of antigenicity at E-126(Glu→Lys). T h e amino acid substitution might be correlated with mouse neurovirulence. Phyloge netic trees showed that D2-43 strain had close relationship with NGC strain, Ta i wan-87 strain and Philippines-83 strain, while D2-04 strain was closely relat ed to Jamaica strain and Brazil-90 strain. This result demonstrated the existence of different DEN-2 virus infection in our country.
