摘  要:目的观察病理细胞核DNA定量与病理形态的关系,为DNA定量直接用于肿瘤临床诊断打下基础。方法用CAS—200型细胞分析系统对眼部表面肿瘤病例40例的石蜡组织行细胞核DNA含量测定,其中鳞癌5例;原位癌7例;局部癌变7例;不典型增生Ⅲ级6例、Ⅱ级4例、Ⅰ级6例;良性增生2例;正常淋巴细胞3例。结果 DNA含量随细胞异形程度的加重而递增,DNA直方图从单峰至多峰、右移和离散;>5C细胞的出现能区分不典型增生Ⅱ级和Ⅲ级;≥8C细胞对诊断癌变、原位癌和鳞癌有决定性意义。结论计算机图像分析技术测定细胞核DNA含量,对肿瘤临床诊断很有价值。Purpose To analysis the relationship between DNA content of pathological nuclei and their pathomorphological study, and use this technique as a tool in tumor' s clinical diagnosis. Methods In 40 cases of external ocular tumor which are kept in paraffin, we use CAS-200 cell analysis system to exam the DNA content of their nuclei.The whole group consists of 5 cases of squamous carcinoma,7 carcinoma in situ, 7 local carcinous changes, 16 dysplasia including 6 grade Ⅲ 4 grade Ⅱ and 6 grade Ⅰ , 2 benign byperplasia, 3 normal lymphocyte. Results The content of DNA rises as the dysplasia increases. DNA histogram changes from single peak to several peaks, with peaks moving right and scattering. The appearance of cell which is > 5C can distinguish dyssplasia grade Ⅱ from grade Ⅲ , while that of the ≥ 8C can be pathognomonic to carcerous change, carcinoma in situ and squamous carcinoma. Conclusions Computerized image analysis of nuclei DNA content is valuable in the clinical diagnosis of tumor.
