摘  要:目的探索实验性变应性脑脊髓炎(experimentalallergicencephalomyelitis,EAE)动物模型的制备,了解其病理改变,为多发性硬化免疫病理机制及实验性治疗的研究提供实验依据。方法采用免疫诱导方法制备EAE模型,利用光镜、电镜观察豚鼠EAE模型的组织学改变。结果至免疫后18d,全部豚鼠均出现EAE临床症状;光镜下见脑实质及蛛网膜下腔小血管充血,血管周围炎性细胞浸润,呈“袖套”状改变。电镜下见髓鞘明显分层、水肿甚至断裂,轴突细胞器消失。结论EAE模型建立方法稳定、可靠;其病理改变为血管周围炎性浸润、白质脱髓鞘。Objective To investigate establishing the model and pathological characteristics of EAE model.In order to provide the experimental value for research on the immunopathology and experimental therapy in MS.Methods Using the method of immune inducing to establish the EAE model.The pathological changes of EAE were studied in guinea pig with the aid of light and electron microscope.Results The model group all showed the clinical symptom. The characteristic light microscopical findings:congestion and inflammatory cuffs of small blood vessels were evident within cerebral parenchyma and subarachnoid cavity. Electron microscopy showed evidence demyelination around blood vessels. Conclusion The method of establishing the EAE model is stable and reliable. EAE is characterized by perivascular inflammatory cell infiltration and demyelination in white matter.
