伪狂犬病病毒上海株的gI基因的克隆和序列分析

摘　　要：参考 Genebank发表的伪狂犬病病毒( Pseudorabies Virus,PRV)的 g I糖蛋白基因序列 ,自行设计合成一对引物 ,对 PRV上海株( PRV-SH)进行 PCR扩增 ,产物经琼脂糖电泳分析 ,呈现一条约 960 bp的条带 ,将其克隆入p GEM-T-easy载体中 ,并进行序列测定 ,与PRV Rice株 g I基因比较发现 ,核苷酸的同源性为 94.7% ,氨基酸的同源性为 91 .3 % ,证实为gAccording to the Genebank open sequence,a pairs of primer was designed in order to amplify gI gene of pseudorabies virus Shanghai strain.The gI gene was obtained by polymerase chain reaction(PCR) ,subsequently cloned into pGEM T vector.The nucleotide and amino acid sequences of SH strain gI gene were compared with the corresponding sequences of Rice strain,the nucleotide and amino acid homology of SH strain with Rice being 94.7% and 91.3% respectively revealed that It is gI gene.
