摘  要:通过对几种产油真菌的培养 ,详细介绍了菌体油脂的观察、检测方法———苏丹黑染色法 ,并以土生假丝酵母和卷枝毛霉 3 2 2 0 8为例进行了前处理方法及油脂提取方法的分析 .结果表明 :苏丹黑染色法中的定性观察法和定量检测法均能成功地适用于酵母菌 ;超声波处理、盐酸处理是较为有效的前处理方法 ,在实际工作中 ,超声波处理和反复冻融结合使用效果好 ;酵母菌及毛菌油脂的有效提取方法为氯仿Several oleaginous fungi were cultured,and the observing and detecting methods-Sudan Black B straining -for intercellular oil granule or drip were discussed in detail.Taking Candida humicola and mucor circinelloids 3.2208 for example,various pretreatment and extraction methods were employed for obtaining internal lipids of these fungi.The results revealed that both qualitative observing and quantitative testing by Sudan Black B straining could be applied for yeast successfully.Ultrasonic wave and hydrochloric acid were the most effective pretreatment methods,while the combination of ultrasonic wave and freeze thaw was more satisfied in practice.As for the extraction of intercelluiar lipids,chloroform methanol was determined as an effective solvent for both yeast and mucor.
