摘  要:采用分子沉积技术制备了α 淀粉酶的自组装膜 ,并用AFM研究了在不同的pH值溶液和不同电荷密度基底上得到的自组装膜 .结果表明 :pH值从 6 .0变到 4.0 ,形状变得尖细 ,膜中缺陷点多 ;基底的电荷密度增大时 ,酶分子的外观形状较细 ,膜中缺陷点少 ;α 淀粉酶自组装膜的厚度保持约 5~ 10nm .It is of great interest to prepare artificial architecture of protein in thin film, which has been the focus of bioengineering in recent years. The self-assembled films of α-amylase on the negatively charged PET(PET-CO 2 -) is fabricated and the AFM results are described in this paper. The surface of PET-CO 2 - was covered by high-density layer of α-amylase, whose thickness was 5~10 nm. The molecules of α-amylase aggregated in the layer which was obtained from the pH 6.0 solution, and the shape of molecules became sharper, which resulted in lager RMS. On the lower-charged substrate (PET-CO 2 -), the aggregating molecules could be found. With raising the charge density of substrate the shape of molecules became sharper.
